Solid Phase Extraction (SPE) procedure:Note: The solvent volumes shown below are for a 60 mg bed mass. The solvent volumes will need to be adjusted for a smaller or larger bed mass.
Load:Pre-treatment: To 2mL of urine add 2mL of 100mM Sodium acetate buffer (pH 5) spiked with 10ng/mL internal standard. Load pre-treated sample directly - no conditioning required
Dry:10min @ >10 in Hg
Final Prep and Analysis: Evaporate eluant to dryness and reconstitute in 1 mL of 20% methanol in water.
Phencyclidine was spiked into the urine at the 25ng/mL cutoff. Inject: 5 µL on HPLC / Mass Spectrometer (MS) @ (500 °C)
