Solid Phase Extraction (SPE) procedure:Note: The solvent volumes shown below are for a 60 mg bed mass. The solvent volumes will need to be adjusted for a smaller or larger bed mass.
Load:Pre-treatment: To each 2 mL urine sample, add 500 uL of concentrated Hydrochloric acid. Heat at 90 °C for 2 hours. Add 2 mL of 200 mM Sodium acetate buffer (pH 4.0). Add 1 mL of 6 N Potassium hydroxide then vortex. Centrifuge for 5 minutes at 5000 rpm. Verify that pH is between 4.0-6.0. Load pre-treated sample directly - no conditioning required
Dry:10 min @ 10" Hg
Final Prep and Analysis: Evaporate eluant to dryness at 50°C. Add 50uL of Propionic anhydride and 50uL of acetonitrile. Heat 30 mins at 60°C. Evaporate to dryness at 50°C. Reconstitute in 100uL of Ethyl Acetate

Inject: 1 µL on GC / Mass Selective (MSD) @ amu / (320 °C)
