Sample pre-treatment
To 200uL Human urine sample add 100uL 0.1M ammonium acetate buffer (pH 4.0 ) in a glass test tube. Mix/vortex another 15 secs. Add 40uL b-Glucoronidase solution (100,000 units/mL from Campbell Science; Product # DR2102). Vortex 10-15 secs.
Incubate for 2 hours in a shaker at 55o C to complete hydrolysis of the glucuronides.
Add 400ul of 0.1% FA (formic acid) followed by vortexing for another 60 secs.
SPE protocol
Condition: 1 mL methanol
Equilibrate: 1 mL water
Load: Sample from pre-treatment step
Wash 1: 1 mL water
Wash2: 1 mL 40% methanol
Dry: 3 to 4 mins at high vacuum (~10-15" of Hg)
Elute: 2x200 uL (2 aliquots of 200 uL) of methanol
Dry Down: Not required
Direct injection: Eluted samples directly shot on the LC-MS-MS |
