Which workflow is the best for your lab? Manual Extraction Workflow

Step
Description (Using rT3 method as an example)
Beads preparation
Wash beads twice and prepare beads suspension in 1:1 (v/v) methanol/water
Sample
Add internal standard to serum sample (200 µL) and mix with 2% formic acid (200 µL)
Bind
Add 100 µL of bead suspension to pre-treated sample, shake for 1min, incubate for 2 min, magnetic separation for 1 min, then discard supernatant
Wash
Wash twice, each time using 500 µL of 2% formic acid, shake for 1min, magnetic separation for 1 min, then discard supernatant
Elute
Elute with 200 µL of methanol, shake for 2 min, magnetic separation for 1 minutes, then collect supernatant

Automated Extraction Workflow

Step
Description (Using rT3 method as an example)
Beads preparation
Wash beads twice and prepare beads suspension in 1:1 (v/v) methanol/water
Bind
Add 100 µL of bead suspension to pre-treated sample, mix by agitating tip-comb for 3 min, then collect the beads and transfer them to wash 1 wells.
Wash 1
Mix the transferred beads with Wash 1 solvent (500 µL of 2% formic acid) by agitating tip-comb for 1 min, then collect the beads and transfer them to wash 2 wells.
Wash 2
Mix the transferred beads with Wash 2 solvent (500 µL of 2% formic acid) by agitating tip-comb for 1 min, then collect the beads and transfer them to elute wells.
Elute
Mix the transferred beads with elute solvent (200 µL of methanol) by agitating tip-comb for 2 min, then collect the beads and transfer them to waste wells.
Waste
Release the transferred beads in waste wells

Comparison Between Manual and Automated Methods for rT3 Analysis in Human Serum

Absolute recovery was calculated by comparing the rT3 peak areas of pre-extraction-spiked samples with those of post-extraction-spiked samples. Matrix effects were assessed by comparing post-spike serum samples with neat standard solutions at the same concentration.

Frequently Asked Questions About Strata-Mag RP

1. What is Strata-Mag RP?

Strata-Mag RP is a reversed-phase magnetic bead sample preparation technology designed for LC-MS workflows. It combines the selectivity of reversed-phase extraction with the simplicity of magnetic separation, enabling efficient sample cleanup and easy integration with automated laboratory workflows. It is particularly well suited for hydrophobic and weakly polar analytes commonly measured in clinical laboratories.

2. Which clinical applications is Strata-Mag RP best suited for?

Strata-Mag RP is optimized for clinical LC-MS applications involving hydrophobic analytes, including reverse triiodothyronine (rT3), steroid hormones. These assays often demand effective matrix cleanup, high reproducibility, and scalable workflows to support growing testing volumes in clinical laboratories.

3. How does Strata-Mag RP compare with traditional SPE?

Unlike traditional SPE methods that rely on vacuum manifolds or positive-pressure systems, Strata-Mag RP uses magnetic separation to simplify extraction workflows. This can reduce hands-on time, minimize solvent consumption, eliminate flow-rate variability, and make automation easier while maintaining the sample cleanliness and performance expected from chromatography-based extraction methods.

4. Can Strata-Mag RP be automated?

Yes. Strata-Mag RP was developed with automation compatibility in mind. The magnetic separation workflow can be incorporated into manual, semi-automated, or fully automated sample preparation platforms, helping laboratories increase throughput while reducing operator-to-operator variability.

5. What benefits does Strata-Mag RP provide to clinical laboratories?

Strata-Mag RP helps clinical laboratories simplify sample preparation while improving operational efficiency. Key benefits include:

  • Reduced hands-on processing time
  • Improved workflow reproducibility
  • Lower dependence on highly skilled operators
  • Easy implementation on automated platforms
  • Lower solvent and consumable usage
  • Scalable processing for high-throughput testing environments

These advantages can help laboratories meet increasing sample volume demand for clinical assays while maintaining high data quality.
