Innovation in Hormone Testing with Phenomenex
Explore the forefront of clinical research with Phenomenex's advanced hormone testing using LC and sample prep methods.

Posted on Jan 9, 2024

With new technology and emerging hormone therapies, hormone testing is more vital than ever for clinical research. Interest in consolidation of steroid methods by combining these compounds into multi-analyte panels is increasing. Fast LC and sample prep methods encompassing expanded groups of analytes helps labs increase efficiency and throughput. Serum is the preferred sample for hormone testing for clinical research applications.

Phenomenex has always been at the forefront of steroid analysis with applications and products to support your clinical research studies. In a new white paper, selectivity on six different LC columns for 19 steroid analytes are detailed.  Separation of all 19 steroid analytes is achieved in just 8 minutes using a core shell C18 column:

One trend in clinical research is using smaller volumes of serum for analysis. However, extraction methods still require high recoveries to achieve desired sensitivity. Many research studies demand very low limits of detection (LOD). This work describes two sample clean-up methods for serum samples, solid phase extraction (SPE) and supported liquid extraction (SLE). Each technique has its own advantages. SLE provides very clean samples with minimal method development and extraction steps. SPE allows modification of wash and elution steps to selectively remove interferences and elute compounds of interest, but it requires more method development and more extraction steps than SLE:

Often, labs desire a more selective or sensitive method for a specific group of steroid analytes. In addition to optimum LC and sample prep methods for a 19-analyte steroid panel, we provide the best LC and sample prep options for select groups of hormones like estrogens, androgens and cortisols.  For example, using a biphenyl column and different LC conditions resolves testosterone from epitestosterone:

SLE extraction of estrogens using Novum Pro with derivatization gives better sensitivity for low level testing by mass spectrometry:

SLE was also a better choice for glucocorticoids. Each one of these subgroups has its own preferred sample preparation and LC conditions. These conditions can be found in one place in this white paper.
