Why do I need to use a syringe filter or filter vial before LC‑MS or HPLC analysis?

Filtering removes particulates that can clog autosampler needles, tubing, and column inlet frits. This clogging could cause back‑pressure increases, retention time shifts, and poor peak shapes. Syringe filters and filter vials provide a fast, consistent way to keep the system clean and extend column lifetime. Using the correct membrane (PTFE, Nylon, RC, and PES, etc.) prevents analyte loss and ensures chemical compatibility.

Which membrane type should I choose for syringe filtration?

Membrane choice depends on solvent compatibility and analyte properties. PTFE is ideal for organic solvents and aggressive chemicals. Nylon and RC (Regenerated Cellulose) work well with aqueous and mixed solvents, while PES is suitable for aqueous biological samples due to low protein binding. PVDF offers broad compatibility for many LC‑MS workflows. Always avoid membranes that may adsorb your analyte.

Why do I need to dilute or acidify samples before protein precipitation?

Protein precipitation works by disrupting protein solubility, causing aggregation and precipitation. Acidic conditions and organic solvents (such as acetonitrile or methanol) help denature proteins more completely. Acidifying the sample improves analyte recovery for basic compounds by preventing their adsorption to proteins. It also improves reproducibility from well to well.

What is Supported Liquid Extraction (SLE), and when should I use it?

SLE mimics traditional liquid–liquid extraction but uses a solid support to immobilize the aqueous phase. This allows easy partitioning into an organic solvent. SLE is excellent for extracting moderately hydrophobic analytes from plasma, urine, or other aqueous sample matrix with minimal emulsions and high reproducibility. It provides cleaner extracts than protein precipitation and has simpler workflows than SPE. Use SLE when you need high recovery, lower matrix effects, and better method robustness.

When should I choose SPE over other sample preparation methods?

SPE offers high selectivity, excellent cleanup, and compatibility with a wide range of analyte classes. Choose SPE when you need maximum removal of matrix interferences, high‑purity extracts, or preconcentration of low‑level analytes. Compared to protein precipitation or SLE, SPE delivers cleaner extracts and lower matrix effects, leading to more sensitive and robust LC‑MS results.
