Vitamins B1 and B6 are two water-soluble vitamins with clinical research interest. Chromatographically, these are very challenging analytes for a reversed phase separation as they have negative values for the octanol/water partition coefficients. Presented here is a simple method that does not involve derivatization or ion pairing reagents. The method involves whole blood extraction at lower pH, cost effective internal standards, no derivatization and a reversed phase LC/MS/MS compatible method for the analysis. The assay is evaluated for precision, accuracy, linear range, and the results meet acceptance criteria.
