Development of a Miniaturized SPE Method for the Extraction of Therapeutic Oligonucleotide in Small Amounts of Matrix Sample
This study showcases the utilization of Clarity OTX 2 mg/well microelution SPE plates for efficient extraction of oligonucleotides (Nusinersen) from minimal serum volumes (10 µL or less). The isolated material facilitates subsequent characterization and quantitative LC-MS/MS testing, demonstrating excellent linearity and sensitivity even at low pg/ml concentrations. The direct injection approach post-SPE elution not only enhances efficiency but also eliminates the requirements for evaporation, reconstitution, and reduces potential user errors.
