FAQs

This is very analyte dependent, if analytes are well retained in the HPLC method, then it is likely that you can inject directly without issues. If analytes elute early in the run, it may be necessary to dilute the sample with water, or a buffer, to weaken the solvent strength of the injection solvent prior to analysis.
Phree is very specific to phospholipids. Analogs of phospholipids, such as long chain fatty acids may also be removed. This is an added benefit for analyses from biological samples. Very few analytes are as hydrophobic as phospholipids so analyte retention on Phree is not a concern. Although there is no method development required to use Phree, it is still important to know what the analytes of interest are to make sure Phree is appropriate for the application.
• Option 1: Osmotic Breakdown-Add 3 parts D.I Water to one part whole blood sample. Mix/vortex for 3-5 seconds Centrifuge at 3700 rpm for 10 min and collect supernatant • Option 2: Sonication-Sonicate whole blood sample for 15 min at room temperature and centrifuge at 3700 rpm for 10 min and collect supernatant • Option 3: Inorganic salts-Add three parts 0.1M Zinc sulfate/0.1M ammonium acetate solution to one part whole blood sample. Mix/Vortex for 3-5 seconds and centrifuge at 3700 rpm for 10 min and collect supernatant.
In order to achieve consistent flow rates when running multiple tubes simultaneously it is advisable to use a vacuum pump which can generate a vacuum equivalent to 20" of mercury.
